چکیده

The role of DNA replication in the synthesis of RNA and proteins during the “late” period of the infective cycle of T!%nfected Escherichia coli F has been examined. Inhibition of DNA synthesis was achieved by addition of chloramphenicol to TS-infected cultures or by use of a mutant of T5 (T5 am I), which is unable to cause the synthesis of an active DNA polymerase upon infection of the nonpermissive host, E. coli F. The synthesis of late or Class III RNA and lysozyme, a presumed late protein, was measured. In the absence of DNA synthesis a small amount of late or Class III RNA species is formed by 9 to 12 min after infection, but much greater quantities are produced under conditions allowing concurrent DNA replication. The synthesis of T5 lysozyme occurs normally in both the permissive and nonpermissive hosts.

Ribonucleic acid synthesis in T2-infected Escherichia coli during "stringent" control.

The leucine auxotroph Escherichia coli 2961 exhibited stringent control of net ribonucleic acid (RNA) synthesis during amino acid starvation. After leucine was exhausted from the medium, the rate of uracil incorporation into RNA rapidly decreased to 2 to 4% of the prestarvation value. Infection of the starved cells with T2 phage stimulated uracil incorporation to a level equivalent to that of u...

Fine-structure mapping of the firA gene, a locus involved in the phenotypic expression of rifampin resistance in Escherichia.

The firA (Ts)200 mutation not only eliminates the resistance to rifampin of certain genetically resistant strains, but, moreover, renders ribonucleic acid synthesis thermolabile. The firA gene has been mapped by P1 tranduction and is located extremely close to the structural gene for deoxyribonucleic acid polymerase III at 4 min on the Escherichia coli linkage map.

Membrane-bound ribonucleic acid synthesis in Escherichia coli infected with ribonucleic acid bacteriophage R23.

1. A cell membrane-DNA-RNA polymerase complex which contains virtually all of the RNA-synthetic capacity of the cell has been isolated from uninfected and R23-infected Escherichia coli. 2. Infection of E. coli by RNA bacteriophage R23 inhibits the RNA synthetic capacity of the membrane complex. In infected cells some of the RNA synthetic capacity is released from the membrane fraction. 3. The m...

Effect of the "ribonucleic acid control" locus in Escherichia coli on T4 bacteriophage-specific ribonucleic acid synthesis.

Amino acid control of ribonucleic acid (RNA) synthesis in bacteria is known to be governed genetically by the rel locus. We investigated whether the rel gene of the host would also exert its effect on the regulation of phage-specific RNA synthesis in T4 phage-infected Escherichia coli cells. Since T-even phage infection completely shuts off host macromolecular synthesis, phage RNA synthesis cou...

Amino acid regulation of messenger ribonucleic acid synthesis in T4-infected Escherichia coli.

The effect of amino acid starvation on the synthesis of T4 messenger ribonucleic acid (mRNA) was studied by measuring the uptake of labeled precursors into Escherichia coli cells which were infected prior to amino acid starvation. The results indicated that the rate of uptake of labeled uracil and adenine into T4 mRNA was reduced by approximately 50% when amino acids were withheld from stringen...
